PREMADE LIBRARY SEQUENCING

Your library, sequenced on AVITI

Send us your finished libraries and our experienced team will sequence them on the Element Biosciences AVITI with Cloudbreak chemistry, from low-output runs to full flow cells.

Read configuration and output are matched to your library and application, with every library checked before loading and data delivered from library receipt in five business days.

SPECIFICATIONS

Sequencing specifications

Read length, depth and output are configured to your library and confirmed in writing at quote. Everything below can be copied directly into a specification or procurement document.

Output here is configured, not ordered from a menu. Read length, depth and reads per run are set against your library and against what the data has to support, and confirmed in writing at quote.

We deliberately do not publish a fixed yield table, for two reasons.

A vendor's published read counts are measured on the vendor's own libraries and will not be what your library returns, so quoting them at you would be borrowing someone else's number to describe your run.

And the figure that matters is the depth your project needs, not the largest one a kit can reach. Tell us the library type, how it was prepared, the read configuration and the depth you are aiming for, and we will tell you what is workable and what it costs before you send anything.

Platform and chemistry

Element Biosciences AVITI, Cloudbreak chemistry

Read configuration

Single-end and paired-end at multiple read lengths. PE50, PE100, PE150 and SE100 are the common choices, and the list is not a limit. Configured to your library and application

Output level

Low, medium and high output, matched to the depth your project needs. 2 × 75 low output is available in the Cloudbreak Freestyle kit configuration only

Reads per run

Set by the configuration and output level agreed for your project, and confirmed at quote. Yield depends on your library type and preparation, so we quote against your library rather than against a published figure

Base quality

Q30+ as standard. Cloudbreak UltraQ delivers greater than Q50 on the majority of reads, available on request

Indexing

UDI and UMI supported, with full demultiplexing. UMI-based error correction on request

Library input

Assessed on the library you have rather than gated by a published minimum. Send us the details and we will confirm before you ship

Library compatibility

Most standard linear and circularised preparations, including TruSeq-style, Nextera-style and NEBNext. IDT and SwiftBio Normalase normalisation chemistry is not compatible

Data delivery

Demultiplexed FASTQ files and quality reports by secure AWS link. No portal login and no file size limits

Turnaround

Five to ten business days from library receipt to data delivery

UDI and UMI support

Unique dual indexing and unique molecular identifiers are fully supported.

Indexed libraries sequenced with full demultiplexing

UMI sequences preserved in read headers for downstream processing

UMI-based error correction performed on request

Error correction is not performed by default, so ask for it at scoping.

how it works

From library receipt to data delivery

1. Library receipt
Pre-prepared libraries received and checked.
2. Pre-run QC
Library concentration and quality assessed before the run. If there is a problem, you hear about it before we sequence anything, so the decision on how to proceed is yours.
3. Sequencing configuration
Flowcell, read configuration and output level selected against your project requirements.
4. Sequencing
On AVITI, using Cloudbreak chemistry.
5. Post-run QC
Run assessed against cluster density, Q30, GC bias, quality distributions and adapter content.
6. Demultiplexing
Indexed libraries demultiplexed and organised by sample.
7. Data delivery
Demultiplexed FASTQ files and quality reports, delivered by secure AWS link. No portal logins and no file size limits.
8. Optional bioinformatics
Analysis matched to your application – from alignment and variant calling to differential expression or a custom pipeline – on request.

Typical turnaround: 5 business days from library receipt to data delivery.

Talk to a scientist about your targets

What you receive

Demultiplexed FASTQ files

Raw reads organised by sample, ready for alignment or downstream analysis, delivered by secure AWS link.

Oligonucleotide-conjugated antibodies capture the target, release it, and capture it again. Each step strips away unbound antibody and non-specific complexes, so the background that limits conventional immunoassays is removed before the signal is ever read.

Quality reports

Per-base quality, GC content, adapter sequences, Q30 metrics and other relevant run-quality measurements.

Beads are loaded into an array of femtolitre-scale wells, each sized to hold a single bead, and sealed.

Detailed run reports

Post-run reporting covering the sequencing and quality metrics that matter for your application.

The same automated platform supports two routes:

  • qPCR for fast single-plex and low-plex quantification of established biomarkers

  • NGS for high-plex discovery across large target sets

Programmes can move from broad discovery to focused validation without changing platform, laboratory or antibody pairs, so the data stays comparable across the life of the study.

Optional bioinformatics

Metagenomic classification, diversity analysis or custom pipelines. Scope is defined per project and is not part of the standard service.

The ARGO HT System automates the NULISA workflow, which keeps turnaround short and precision steady across plates.

Cohorts of thousands, and collections that run for years, depend on that.

QUALITY CONTROL

Quality control before, during and after the run

Every library is checked before it goes on the sequencer, monitored while the run is in progress and assessed again before data is released.

Pre-run QC

Every library is checked for concentration and quality before loading. If we find an issue, we tell you before sequencing starts so you can decide whether to proceed – sequencing a library we already know is compromised wastes your budget and your material.

During-run QC (new card – the heading promises “during” but there are only two cards; if you don’t want a third, drop “during” from the heading)

Run metrics are monitored while the run is in progress, so a problem is caught early rather than discovered once the data is delivered.

Post-run QC

Once the run completes, we check cluster density, Q30, GC bias, quality score distributions and adapter content.

Every dataset is delivered with a detailed run report.
